Pharmacology :
Ivermectin is an avermectin macrocyclic lactone. It interacts with glutamate-gated chloride ion channels. By increasing membrane permeability to chloride ions it causes irreversible neuromuscular blockade in nematodes and arthropods leading to paralysis and death. Beside its effectiveness against ticks, mites and insects ivermectin is a broad spectrum anthelmintic effective against mature and immature stages of nematodes, including inhibited stages and strains resistant to other anthelmintic classes.
Triclabendazole is a benzimidazole. It binds to tubulin impairing intracellular transport mechanisms and interferes with protein synthesis. Triclabendazole is specifically effective against all 3 stages of Fasciola. The unique efficacy against early immature and immature stages makes
VERCET particularly useful for the treatment of outbreaks of acute and subacute fasciolosis and for the strategic control of Fasciola infections.

Triclabendazole is active against liver fluke, however unlike all the other benzimidazole, triclabendazole has no activity roundworm

In cattle & sheep triclabendazole at 10 mg/kg, PO, is highly effective against immature Fasicola hepatica in the liver parenchyma and agaist the mature stages in the bile ducts.

The maximum tolerated dose in the target spices is 200 mg/kg thus the safety margin is 20 times the therapeutic dose.
